Testing vocabulary would mean that the examiners want to know how good your knowledge is about the words in English. This does not only include their meanings but also their opposites, usage and grammar. So if you merely know the meaning of a word, it is not enough.
You must have complete knowledge about how that word is used in English.
English is a universal language; people all around the world use it for communicating. Without this language it would be impossible for people of different nationalities to come together. Moreover it is the medium of instruction in colleges in a number of countries. Thus it is important for you to be well versed with the language. After all there is no point in taking admission in a college where you can neither understand the lectures nor write good answers in the exams. The colleges also want students to know English well so that they can teach efficiently and obtain good results. Thus it is better for the test takers, as well as you, to test how well you understand English. The exam like GRE/TOEFL/IELTS does this job of scanning students for their language proficiency. The test contains a wide variety of questions which test your language skills in a comprehensive manner.
Vocab Zone:
The vocab zone here measures your ability to analyze information and interpret it. It tests whether you can analyze relationships between different words.
Words (Synonyms and Antonyms):
Your knowledge of the words in English language is of great importance in test. This testing of your words is comprehensive and also very important for scoring. Mere superficial knowledge of the meanings is not sufficient to score. The test is designed such that you will have to have complete knowledge about the meanings, the antonyms and the usage of the words. You shall be tested for antonyms, analogies, comprehension etc. Hence it is not enough to know the meanings but you must also know how to use the words.

GRE tests, besides other things, measure your vocabulary. The tests are designed such that they measure your knowledge of words in direct and indirect ways. The verbal section is devoted to test how well you understand the meaning of the words of English and the analytical writing section tests the way you use words in an indirect way. You exhibit your knowledge of words here by writing independently without any restriction. So it is more important to concentrate on the verbal section for vocabulary. You need to prepare separately for this section
Each language is a collection of words threaded in sentences using the rules of grammar. It is these sentences which give us the capability to interact with each other; they form the base for communication. If two persons do not know the same language it becomes very difficult for them to communicate. And if these two people happen to be a teacher and a student then the process of teaching and learning is impossible. The medium of instruction has to be understood by the student. So when you go to a college to study it is expected of you to know the language of instruction well. Now, to know a language well does not just mean that you are able to speak the language but also that you can read and write it well, that you can understand what the other person is saying and that you can express. Hence you need to have a good command over the grammar and understand a wide variety of words. Your vocabulary should be of the level of the teachers of the college teaching you. Moreover you should be able to understand a number of different subjects. Then only is the process of teaching and learning successful. Thus it becomes obvious that your vocabulary is tested in GRE.
